چکیده
conclusions these results show that zen and α-zol enhance the rate of cell division in er positive cells and therefore, exposure to this mycotoxin may increase the risk of breast cancer. results the results obtained from the present study demonstrated that both zen and α-zol enhance the cell viability of mcf-7 especially at low doses (1 - 500 ng/ml) and at a high dose of 125µg/ml after 24 and 48 hours. however, this effect for α-zol was somewhat greater than that for zen. on the other hand, these estrogenic compounds did not have any effect on the cell viability of mda-mb-468. no morphological change was observed in treated cells. objectives the present study was designed to investigate the effects of zen and α-zol on human breast cancer cell lines mcf-7 and mda-mb-468. materials and methods cell lines were treated by low and high doses of zen and α-zol (0, 1, 30, 62, 125, 250 and 500 ng/ml and 1, 2, 4, 8, 15, 30, 62 and 125 µg/ml) for 24 and 48 hours. then mtt colorimetric assay was used to evaluate the cytotoxicity effect of zen and α-zol. furthermore, morphological changes of treated and untreated cell lines were studied under an inverted microscope. background in the field of human breast cancer, the most recent researches referred to the influence of endogenous estrogen or exposure to environmental estrogen, as risk factors. zearalenone (zen) as a mycotoxin and its derivative α- zearalenol (α-zol) are known nonsteroidal estrogenic compounds with potential endocrine disrupting properties.

Cell Kinetic Study of Tamoxifen Treated MCF-7 and MDA-MB 468 Breast Cancer Cell Lines
Apoptosis could be a major mechanism of antitumor effect of tamoxifen. Therefore this study is designed to characterize the kinetic behavior of tamoxifen-induced apoptosis in the estrogen receptor positive (ER+) and negative (ER-) cell lines, MCF-7 and MDA-MB-468. Frequency of cell death was examined by trypan blue and acridine orange staining. Annexin V-Fluorescein/PI was used in flow cytometr...

متن کاملApoptotic potential of two Caryophyllaceae species in MCF-7 and MDA-MB-468 cell lines
Background and objectives: Plants have been used to treat diseases like cancer for many years and today the trend towards their use is increasing. One of the most effective mechanisms of plants against cancer is inducing apoptosis. Apoptosis is a programmed cell death which acts opposite to cell division. It starts in response to some stimuli. Despite the effectiveness of apopt...

متن کاملDefective repair of 8-hydroxyguanine in mitochondria of MCF-7 and MDA-MB-468 human breast cancer cell lines.
Breast cancer is one of the major causes of mortality among women in the United States. Although the causes of breast cancer remain unclear, it has been speculated that DNA base damage may lead to mutations that subsequently can be carcinogenic. Recently, defective oxidative DNA damage repair has been implicated in breast tumorigenesis. The major oxidative DNA lesion, 8-hydroxyguanine (8-oxoG),...
متن کاملthe effect of rapamycin on oxidative stress in mcf-7 and mda mb-231 human breast cancer cell lines
conclusions these outcomes support our theory that rapamycin increases oxidative stress in mcf-7 and mda mb-231 cells but also shows high levels of antioxidant effects, which probably limit the effects of the rapamycin on the same issue in the clinic. methods the mcf-7 and mda mb-231 cell lines were cultivated and treated with rapamycin for 72 hours. the viability of the cells was determined us...
